摘要

C-C 基序趋化因子受体 8(C-C motif chemokine receptor 8,CCR8)是一种调控免疫细胞趋化迁移的7次跨膜蛋白,在肿瘤微环境中高表达于Treg表面。由于CCR8+ Treg能强烈抑制肿瘤免疫,因此CCR8是一种扭转抑制型肿瘤微环境的理想靶点。目前有多种在研的靶向CCR8抗体药物,可在多种肿瘤模型中特异性清除肿瘤浸润CCR8+ Treg、扭转免疫抑制,展现出良好的抗肿瘤活性。此文对CCR8+ Treg促进肿瘤进展的基础研究和靶向药物应用进行综述,以期为该靶点肿瘤免疫治疗研究提供参考。

Abstract

C-C motif chemokine receptor 8(CCR8) is a kind of seven-transmembrane protein that regulates the chemotaxis of immune cells, and is highly expressed on the surface of Tregs in the tumor microenvironment (TME). CCR8+ Tregs strongly inhibit tumor immunity, so CCR8 is an ideal target for reversing suppressing TME. At present, there are a variety of anti-CCR8 antibody drugs with prominent anti-tumor activity under development, which can specifically eliminate tumor-infiltrating CCR8+ Tregs and reverse immunosuppression in many tumor models. This review summarizes the basic researches on CCR8+ Tregs promoting tumor progression and the application of targeted drugs, in order to provide a reference for tumor immunotherapy study.
