目的 探索一种简便、快捷、准确检测自动移液工作站移液性能的方法。方法 用校准后的移液器移取系列体积的重铬酸钾原液,配制成等体积不同浓度酶标检测稀释液,读取吸光度值,拟合原液体积-稀释液吸光度值回归方程。选取实验室最常用移液量50 μl作为测试点,用96通道自动移液工作站移取50 μl重铬酸钾原液配制稀释液,读取吸光度值,根据回归方程计算出各通道实际原液移取体积,对自动移液工作站的移液准确性、重复性和通道一致性进行分析。结果 原液体积与稀释液吸光度值成线性关系,相关系数为1.000。在50 μl量程点,96个通道的准确性均符合偏差不超出±3%的标准,7次重复性实验结果符合变异系数≤1.5%的标准,一致性符合不超出设定量×(100±5)%的标准。结论 建立的方法简便、快捷、准确,可用于实验室自动移液工作站移液性能的日常监测。
Objective To explore a simple, fast, and accurate method to test the performance of automatic pipetting working station. Methods A series of dilutions of potassium dichromate with different concentrations and same volume were prepared with potassium dichromate original solution of different volumes by calibration pipette. The absorbance (A) value was tested and the regression equation of original potassium dichromate volume and A value of dilutions was fitted. The most commonly used pipette volume 50 μl was selected as a testing point. Potassium dichromate original solution of 50 μl was transferred by automatic pipetting working station with 96 channels, and the A value was tested. The volume of original solution pipetted by each channel was calculated according to the regression equation. Accuracy, repeatability, and the pipetting consistence of channels for the automatic pipetting working station were analyzed. Results The volume of original solution and the A value showed linear correlation, and the correlation coefficient was 1.000. At the point of 50 μl, the accuracy met the standard of deviation within ±3% among 96 channels. The repeatability met the standard of coefficient of variation ≤1.5%. The consistence met the standard of within(100±5)% of set amount. Conclusion The established method is simple, fast and accurate, and can be used as a regular method for routine monitoring of the performance of automatic pipetting working station.
