醛化鹅红细胞用于百日咳菌液血凝滴度测定的可行性研究

摘要

目的  初步评价醛化鹅红细胞用于百日咳菌液血凝滴度测定的可行性,并确定其使用有效期。方法 以新鲜鹅红细胞作为对照,筛选出测定百日咳菌液血凝滴度的最佳醛化鹅红细胞浓度。用3批醛化鹅红细胞分别检测3批百日咳菌液血凝滴度,考察与新鲜鹅红细胞检测结果的一致性。观察不同放置期的醛化鹅红细胞发生溶血的情况,并考察血凝滴度检测结果与新鲜鹅红细胞的一致性。 结果  用于百日咳菌液血凝滴度测定的最佳醛化鹅红细胞浓度为0.33%。不同批次醛化鹅红细胞检测百日咳菌液血凝滴度的结果均与新鲜鹅红细胞检测结果一致。醛化鹅红细胞在2~8 ℃存放90 d仍能准确检测百日咳菌液血凝滴度,120 d后发生溶血现象。结论  0.33%的醛化鹅红细胞可用于百日咳菌液血凝滴度测定,醛化鹅红细胞使用有效期为90 d。

Abstract

Objective  To primarily evaluate the feasibility of using hydroformylated goose red blood cells (RBCs) in the determination of hemagglutination (HA) titers of Bordetella pertussis fluid, and to determine the period of validity. Methods  The optimal working concentration of hydroformylated goose RBCs in HA test was screened using fresh goose RBCs as control. HA titers of 3 batches of Bordetella pertussis fluid were tested by 3 batches of hydroformylated goose RBCs respectively, and results were compared with those of fresh goose RBCs. The hemolysis of hydroformylated goose RBCs stored for different periods was observed and HA test results were compared with those of fresh goose RBCs. Results  The optimal concentration of hydroformylated goose RBCs in the HA test of Bordetella pertussis fluid was 0.33%. HA titers of Bordetella pertussis fluid measured with different batches of hydroformylated goose RBCs were consistent with the results by fresh goose RBCs. HA titers of Bordetella pertussis fluid were still accurately detected using hydroformylated goose RBCs stored at 2-8 °C for 90 d, but hemolysis occurred after 120 d. Conclusion  The 0.33% hydroformylated goose RBCs are feasible for HA test of Bordetella pertussis fluid and the period of validity is 90 d.
