摘要

目的 比较猪源胰蛋白酶与重组胰蛋白酶对鸡胚细胞的消化效果,筛选出可替代猪源胰蛋白酶消化鸡胚细胞的最优方案。方法 对照组用质量百分数0.125%猪源胰蛋白酶溶液,实验组分别用质量百分数0.1%、0.2%、0.3%重组胰蛋白酶溶液按照验证参数消化鸡胚细胞15、20、25 min,进行细胞计数,并监测细胞活率。筛选出最优方案,制备单次病毒收获液,并与0.125%猪源胰蛋白酶对比滴度结果。结果 对照组平均活细胞密度为1.12×106 ml-1,平均细胞活率为95%;实验组最优鸡胚细胞消化条件为0.2%重组胰蛋白酶溶液消化20 min,平均活细胞密度为2.24×106 ml-1,平均细胞活率为97%。2组消化鸡胚细胞效果差异有统计学意义,t=19.65,P<0.05。0.125%猪源胰蛋白酶组病毒滴度为(6.56±0.12)lgCCID50/ml,0.2%重组胰蛋白酶组为(6.89±0.10)lgCCID50/ml, t=6.78,P<0.05,病毒滴度结果差异有统计学意义。结论 以0.2%重组胰蛋白酶溶液消化20 min作为最优鸡胚细胞消化条件,可替代0.125%猪源胰蛋白酶溶液,为今后生产工艺分析研究及清真认证提供了数据和参考。

Abstract

Objective To compare the digestion effect of porcine trypsin and recombinant trypsin in chicken embryo cells, and select the optimal alternative that could replace porcine trypsin in chicken embryo. Method The control group was treated with 0.125% porcine trypsin solution, while the experimental group was treated with 0.1%, 0.2%, and 0.3% recombinant trypsin solution for 15, 20, and 25 min according to the verification parameters for chick embryo cells digestion respectively. The cells were counted and cell viability was monitored. The optimal alternative was selected and used to prepare a single virus harvest solution, and the titers were compared with that by 0.125% porcine trypsin. Result In the control group, the average living cell density was 1.12×106 ml-1, and the average cell viability was 95%. In the experimental group, 0.2% recombinant trypsin solution and digestion time of 20 min was the optimal digestion conditions for chicken embryo cells, with the average living cell density 2.24×106 ml-1, and the average cell viability 97.00%. There were statistically significant differences between digestion effects of2 groups, with t=19.65, P<0.05. The titer of 0.125% porcine trypsin group was (6.56±0.12) lgCCID50/ml, and the titer of 0.2% recombinant trypsin group was (6.89±0.10) lgCCID50/ml, with t=6.78, P<0.05, which showed statistical significance. Conclusion The 0.2% recombinant trypsin solution with digestion time of 20 min can replace 0.125% porcine trypsin solution as the optimal digestion condition for chicken embryo cells, which provides data and reference for production process analysis and Halal certification in the future.
