摘要

目的  探讨Vero细胞洗换方式对口服六价轮状病毒活疫苗收获液的影响。方法  采用3种不同方式洗换Vero细胞后,观察相应的致细胞病变效应,使用ELISA检测洗换前后牛血清白蛋白(bovine serum albumin,BSA)残留、快速荧光灶法检测收获液病毒滴度并测定病毒收获液浊度及粒径,比较3种洗换方式的差异。结果  使用方式123洗换后,方式12致细胞病变进程明显较方式3收获液BSA残留均值±标准差分别为(15.0±4.1)(16.3±4.0)(52.6±4.7) ng/ml,方式12较方式3收获液BSA残留明显下降方式123洗换的收获液病毒滴度分别为(7.27±0.06)(7.23±0.06)(6.97±0.06) lg荧光灶单位/ml方式12较方式3收获液病毒滴度明显上升。不同洗换方式处理后,病毒收获液浊度和粒径的差异无统计学意义。方式123处理后,病毒收获液BSA残留、病毒滴度、浊度与粒径均符合标准。结论  结合实际生产条件,滴度较高、BSA含量较低的方式1适合作为后续的大规模原液制备过程中的洗换方式。

Abstract

Objective  To discuss the influence on virus harvest of oral hexavalent live rotavirus vaccine with different Vero cell rinse ways. Methods  After 3 rinse ways, cytopathic effect was observed. Bovine serum albumin (BSA) residue was tested by ELISA before and after rinse. Virus harvest titer was tested by rapid fluorescence focus, and turbidity and particle size were measured. The difference was compared among 3 groups. Results  The cytopathic effects of ways 1 and 2 were faster than that of way 3. The BSA residues of supernatent after rinse ways 1, 2, 3 were (15.0+4.1), (16.3±4.0) and (52.6±4.7) ng/ml, respectively. The BSA residues in supernatent of ways 1 and 2 were significantly lower than that in way 3. The virus harvest titers of ways 1, 2 and 3 were (7.27±0.06) , (7.23±0.06) and (6.97±0.06) lg fluorescence focus unit/ml, respectively. The virus harvest titers of ways 1 and 2 were significantly higher than that of way 3. There was no statistically significant difference between turbidity for virus harvest before and after 3 rinse ways, as well as particle size. The BSA residue, titer, turbidity and particle size of the virus harvest were all in line with the standard after treatment with ways 1, 2 and 3. Conclusion  The rinse way 1 with higher titer and lower BSA content is more suitable for rotavirus preparation in large-scale bulk based on above studies and production conditions.
