Effect of different conditions on the mRNA content detection in mRNA vaccines by RiboGreen method

目的 探讨RiboGreen法检测mRNA疫苗中mRNA含量可能的影响因素。方法 在不同裂解液浓度、裂解时间、样品稀释浓度和参比品条件下,使用RiboGreen荧光染料法对不同生产工艺的mRNA疫苗的mRNA含量进行检测。结果 在裂解液为0.1%〜2.5% Triton X-100、裂解5〜30 min、样品稀释至mRNA含量0.5〜1.5 μg/ml时,mRNA含量检测结果的差异无统计学意义;不同参比品序列下样品mRNA含量检测结果差异有统计学意义。结论 根据检测样品的mRNA含量的回收率,初步确定了mRNA疫苗中mRNA含量检测的最适条件。

Objecive To investigate potential influencing factors on the mRNA content detection in mRNA vaccines by RiboGreen method. Methods The mRNA contents of mRNA vaccines were detected and analyzed using the RiboGreen fluorescent dye under different lysis buffer concentrations, lysis time, sample dilutions and reference standards. Results There was no statistically significant difference in the results of mRNA content when samples were diluted to 0.5-1.5 μg/ml and lysed in 0.1%-2.5% Triton X-100 lysis buffer for 5-30 min. There was statistically significant difference in the results of mRNA content with different reference standards. Conclusion Based on the recovery of mRNA contents, the conditions are determined initiatively for mRNA content detection in mRNA vaccines.
