摘要

 目的  探索以猪血为原料制备凝血因子ⅩⅢ的新方法。方法  采用硫酸铵分级沉淀法从猪血中粗提凝血因子ⅩⅢ,进一步用二乙氨乙基-Sepharose阴离子交换层析、Sephacryl S-200凝胶层析进行纯化。结果  获得比活为204.4 IU/mg的高纯度凝血因子ⅩⅢ制剂,其收率为53.5%,纯化倍数21.7倍。结论  从猪血中提取的粗制品再经过离子交换层析和凝胶层析进行分离纯化可得到高纯度凝血因子ⅩⅢ。

Abstract

 Objective  To explore a novel method of isolation and purification of blood coagulation factor ⅩⅢ (FⅩⅢ) from porcine plasma. Methods  Crude FⅩⅢ was obtained by ammonium sulfate precipitation from porcine plasma. Purification was carried out first through diethylaminoethyl-Sepharose anion-exchange resin followed by Sephacryl S-200 gel chromatography. Results  The final purified FⅩⅢ specific activity was 204.4 IU/mg, which was 21.7 fold purer and had 53.5% recovery rate. Conclusion  Highly purified FⅩⅢ can be obtained by anion-exchange resin and gel chromatography from porcine plasma.
