摘要

目的  制备2型肺炎链球菌多糖(Streptococcus pneumoniae type 2 polysaccharide,SPNPS2)-破伤风类毒素(tetanus toxoid,TT)结合疫苗(SPNPS2-TT),并研究其免疫原性。 方法  采用溴化氰(CNBr)活化SPNPS2,己二酰肼(ADH)作连接剂,碳二亚胺(EDAC)为耦联剂,将活化的SPNPS2与TT耦联制SPNPS2-TT。以SPNPS2-TT或单纯SPNPS2免疫Balb/c小鼠,用ELISA检测小鼠血清抗SPNPS2 IgG水平。 结果  SPNPS2-TT的各项指标均达到质控标准。SPNPS2-TT免疫小鼠产生的特异性抗SPNPS2 IgG水平显著高于单纯SPNPS2免疫小鼠,且SPNPS2-TT诱生了免疫记忆应答。 结论  采用此项技术制备SPNPS2-TT疫苗是可行的。

Abstract

Objective  To prepare Streptococcus pneumoniae type 2 polysaccharide(SPNPS2)-tetanus toxoid(TT) conjugate vaccine (SPNPS2-TT) and study its immunogenicity.  Methods  The SPNPS2 was activated by cyanogen bromide(CNBr).  Using adipic acid dihydrazide as linking agent,the SPNPS2-TT was prepared by carbodiimide(EDAC)-mediated coupling of SPNPS2 with TT.The Balb/c mice were immunized with SPNPS2-TT or pure SPNPS2, and the anti-SPNPS2 antibodies were detected by ELISA.  Results  All the quality indexes of SPNPS2-TT meet the standards of quality control. The specific IgG antibody levels elicited in the mice immunized with SPNPS2-TT were significantly higher than those elicited in the mice immunized with pure SPNPS2, and only SPNPS2-TT induced immune memory.  Conclusion  The SPNPS2-TT was successfully prepared with feasible technology.
